TEM (Transmission Electron Microscopy)
This method describes general TEM analysis of protein samples with negative staining.
Procedure
- Thaw the sample.
- Apply 10 ul of the sample onto a carbon filmed grid with the excess liquid being removed by blotting a piece of filter paper after 1 minute.
- Apply 10 ul of dH2O onto the grid and remove the excess liquid with filter paper.
- Apply 10 ul of 2% Uranyl Acetate onto the grid and remove the excess liquid with filter paper after 30 seconds.
- Apply another 10 ul of 2% Uranyl Acetate onto the grid and remove the excess liquid with filter paper after 1 minute.
- Allow the grid air dry for a few minutes and store it in a grid box.
- The specimen is examined and photographed on Thermo Scientific Talos L120C transmission electron microscope at an accelerating voltage of 120Kv.
